ArcheoGen Lab

The research and analysis of ancient biomolecules take place in highly specialized laboratories dedicated exclusively to these purposes.


DNA and other biomolecules (proteins, lipids) have the ability to persist for thousands of years under suitable conditions, but they undergo degradation over time, leading to irreversible damage at all molecular levels. Due to this damage, ancient DNA is far less amenable to analysis compared to modern DNA, and any contamination with modern DNA significantly complicates the analytical process. For this reason, a strict system of anti-contamination measures is mandatory for all laboratories working with ancient DNA (aDNA).


The ArcheoGen laboratories are divided into two physically separate facilities. The sampling laboratories are located in the basement of the Department of Archaeology and Museology (Faculty of Art, Masaryk University, Brno). The molecular genetic laboratories are part of the MUNI BioPharma Hub research infrastructure situated at the Bohunice University Campus.


 

ArcheoGen at Department of Archaeology and Museology

In the basement of the Department of Archaeology and Museology at Masaryk University, specialized laboratories for pre-PCR sample analysis and storage facilities are located. Access to these areas is restricted to experts from the ArcheoGen team.

In the pre-PCR laboratories, incoming findings are documented through photography, cleaned, described, and subjected to decontamination processes. For ancient DNA (aDNA) analysis, teeth or petrous bones are typically sampled. After repeated treatment with UVC radiation, the samples undergo detailed photographic documentation. Subsequently, the tissue is ground into fine powder using a drill and aliquoted into microtubes, preparing the sample for the molecular biology analysis.

ArcheoGen in University Campus Bohunice

The ArcheoGen laboratories will become part of Masaryk University's new infrastructure, the MUNI BioPharma Hub, which is being developed to connect research and education. The BioPharma Hub is located at the Bohunice University Campus.

The newly established ArcheoGen laboratories will provide facilities for molecular biological analyses of ancient DNA, including DNA extraction, shotgun sequencing, whole-genome sequencing, as well as data collection and storage for bioinformatic analysis. Construction is expected to be completed by April 2026, with operations and educational activities scheduled to begin in the fall of 2026.
